The polybrene test is intended to be used to determine the cross-matching suitability between a donor’s red blood cell to the antibody present in the patient’s EDTA plasma.

Infung Logo The manual polybrene test is a rapid and generally more sensitive method for the detection of clinically significant Red Blood Cell (RBC) antibodies than conventional methods. Polybrene, a positively charged polymer, is utilized to induce agglutination of RBCs. The non-specific antibodies agglutination is reversed by the addition of the re-suspending solution, while the specific agglutination due to antibody-antigen bridging remains. This method has been proven to be highly sensitive and is widely used in the detection of both complete and incomplete antibodies.

 

Technical Data

The first step of this test involves the addition of a low ionic strength medium (LIM) to the RBC-EDTA plasma mixture (i.e. donor / patient) to facilitate rapid antibody uptake. In the second step, polybrene is utilized to induce agglutination of red blood cells. The last step involves the addition of re-suspending solution. The non-specific agglutination is reversed by the addition of re-suspending solution, while the specific agglutination due to antibody-antigen bridging remains.
